General description

1HAEo- is a human airway epithelial cell line that was immortalized with the origin-of-replication defective SV40 plasmid (pSVori-). The cell line retains characteristic features of normal differentiated airway epithelial cells including a cobblestone morphology, cytokeratin expression, the ability to form tight junctions and directional ion transport. It is unknown whether the donor had cystic fibrosis or not, however the cells do not contain the ΔF508 mutation in the CFTR gene or other CFTR mutations presenting 98% of all CF mutations (1). CFTR sequence analysis revealed only wild-type CFTR sequences, supporting the conclusion that 1HAEo- are derived from normal (i.e. non-cystic fibrosis) tissue (1).

This product is intended for sale and sold solely to academic institutions for internal academic research use per the terms of the “Academic Use Agreement” as detailed in the product documentation. For information regarding any other use, please contact licensing@emdmillipore.com.

Quality

• Each vial contains ≥ 1X10⁶ viable cells.
• Cells are tested by PCR and are negative for HPV-16, HPV-18, Hepatitis A, C, and HIV-1 & 2 viruses as assessed by a Human Essential CLEAR panel by Charles River Animal Diagnostic Services.
• Cells are negative for mycoplasma contamination.
• Each lot of cells is genotyped by STR analysis to verify the unique identity of the cell line.

Storage and Stability

Store in liquid nitrogen. The cells can be cultured for at least 10 passages after initial thawing without significantly affecting the cell marker expression and functionality.

Human papillomavirus (HPV) is now recognised as a major aetiological agent in the pathogenesis of oropharyngeal carcinoma (OPC). HPV-positive tumours are associated with better outcomes compared to HPV-negative tumours, possibly due to differences in their aetiology and/or the tumour microenvironment.
