General description

Desmoglein-3 (DSG3) is a calcium binding membrane protein that is localized desmosome cellular junctions and interacts with plaque proteins and intermediate filaments at cell-cell adhesion points. Desmosomes are cell-cell junctions between epithelial, myocardial and other cells types. Strong intercellular adhesion is critical for tissues that experience mechanical stress, such as the skin and heart. Desmosomes provide adhesive strength to tissues by anchoring desmosomal cadherins of neighboring cells to the intermediate filament cytoskeleton. It is thought that raft membrane microdomains play a role in desmosome homeostasis by regulating the dynamics of desmosome assembly and disassembly. In human keratinocytes, Desmoglein-3 (DSG3) is raft associated and disruption of rafts prevents desmosome assembly. Desmoglein-3 is one of four sister proteins in the desmoglein family. Desmoglein-3 is also the autoantigen for Pemphigus vulgaris (PV) a lethal skin disease that is a result of autoantibodies against Desmoglein-3.

Within stratified squamous epithelia, such as the epidermis, desmogleins are generally expressed in a differentiation-specific manner.
