Application

Target description

Tumor necrosis factor receptor superfamily member 1B (UniProt: P25119; also known as Tumor necrosis factor receptor 2, TNF-R2, Tumor necrosis factor receptor type II, TNF-RII, TNFR-II, p75, p80 TNF-alpha receptor, CD120b) is encoded by the Tnfrsf1b (also known as Tnfr-2, Tnfr2) gene (Gene ID: 21938) in murine species. TNFR2 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 23-258), a transmembrane domain (aa 259-288), and a cytoplasmic domain (aa 289-474). It serves as a high-affinity receptor for TNF- . It displays about five-fold reduced affinity for lymphotoxin- . It acts as a co-stimulator for IL-2 Induction and T cell survival and enhances the responses of lymphocytes to TCR-mediated signaling. It plays a critical role during TCR/CD28-mediated stimulation toward IL-2 induction, clonal expansion, and survival. TNFR2 deficient T cells possess a profound defect in IL-2 production in response to TCR/CD28-mediated stimulation. It is also reported to induce proliferative responses in certain cells of hematopoietic origin and enhances expression of certain adhesion molecules such as ICAM-1. (Ref.: Chen, X., et al. (2010). J.
