Specificity
Clone 5I2-K is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Chromobox protein homolog 5 (CBX5). It targets and epitope within 15 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Chromobox protein homolog 5 (CBX5).
Application
Quality Control Testing
Evaluated by Western Blotting in A431 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CBX5 in A431 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CBX5 in U2OS cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CBX5 in Human gallbladder tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CBX5 in U2OS cells.

Target description
Chromobox protein homolog 5 (UniProt: P45973; also known as Antigen p25, Heterochromatin protein 1 homolog alpha, HP1 alpha) is encoded by the CBX5 (also known as HP1A) gene (gene ID: 23468) in human. Chromobox protein homolog 5 (CBX5) is a component of heterochromatin that recognizes and binds histone H3 tails methylated at lysine 9 (H3K9me), leading to epigenetic repression. It is excluded from chromatin when tyrosine 41 of histone H3 is phosphorylated (H3Y41ph) by JAK2. It can also interact with lamin-B receptor (LBR) and this interaction can contribute to the association of the heterochromatin with the inner NuCLEAR™ membrane. It has a single N-terminal Chromo domain (aa 20-78) that can bind to histone proteins via methylated lysine residues, and a C-terminal Chromo shadow-domain (aa 121-179), which is responsible for the homodimerization and interaction with a number of chromatin-associated non-histone proteins. CBX5 is a suppressor of metastasis, and it is down-regulated at the transcriptional and protein level in metastatic compared to non-metastatic breast cancers. In breast cancer, its expression level correlates with both clinical outcome in terms of patient survival and clinical data in terms of tumor size and stage of this disease. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
