Cell Line Description

This cell line is a derivative of the normal human prostate cell line PNT2 (Sigma cat no. 95012613) adapted to grow in serum-free medium. PNT2 was established by immortalisation of normal adult prostatic epithelial cells by transfection with a plasmid containing SV40 genome with a defective replication origin. The primary culture was obtained from a prostate of a 33 year old male at post mortem. PNT2 cells contain the SV40 genome and express large T protein.

Culture Medium

Defined Keratinocyte-SFM (Gibco 10785) + Growth Supplement (Gibco 10784-015).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:10 i.e. seeding at 2-4 x 10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. Use Trypsin Inhibitor to inactivate the trypsin. When freezing cells down use conditioned medium plus 10% DMSO.
