General description

Importin β comprises 19 tandemly repeated Huntingtin, elongation factor 3 (EF3), protein phosphatase 2A (PP2A), and the yeast kinase serine/threonine-protein kinase-TOR1 (HEAT) motifs and wraps around the importin-β (IBB). It also consists of an N-terminal RAN guanosine triphosphate (GTP) binding domain, a middle zone interacting with the nuclear pore, and an importin α binding domain at the C-terminus. The importin β gene is mapped to human chromosome location 17q21.32.
Monoclonal Anti-Importin b (mouse IgG1 isotype) is derived from the 31H4 hybridoma produced by the fusion of mouse myeloma cells (NS1 cells) and splenocytes from BALB/c mice immunized with importin. Importin β (Imp β), also referred to as karyopherin β1, is an import receptor that belongs to the family of nuclear transport receptors.

Biochem/physiol Actions

Importin β (Impβ) forms a complex with importin α, which, in turn, binds the cargo protein via its NLS (nuclear localization signals). The cargo protein is transported by this ternary complex to the cytoplasmic face of the nuclear pore complex (NPC) via a direct interaction with specific nucleoporins. When the complex reaches the nuclear site of the nuclear pore complex (NPC), Ran-GTP binds to Impβ to form an Impβ/Ran-GTP complex and releases Impα and the cargo protein. The Impβ/Ran-GTP complex is then transported back to the cytoplasm, where the complex dissociates upon hydrolysis of GTP to GDP. Importin β is then ready to participate in new nuclear import cycle.
Importin β regulates cellular functions, such as mitotic spindle assembly and centrosome dynamics. It is implicated in the insertion of integral membrane proteins into the inner nuclear membrane. Importin β also functions as a chaperone for highly basic proteins in the cytoplasm.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
