General description

The Expand High FidelityPLUS PCR System is a blend of Taq DNA Polymerase and a thermostable proofreading protein that lacks polymerase activity.
This enzyme blend is designed to amplify fragments up to 5 kb from all types of DNA with high yield and fidelity.
In addition, this enzyme blend also incorporates dUTP and, in combination with Uracil-DNA Glycosylase, can be used to safeguard PCR reactions from cross contamination.
The fidelity of DNA synthesis with the Expand High FidelityPLUS PCR System shows a 2-fold increase compared to the fidelity obtained with the Expand High Fidelity PCR System and a 6-fold increase compared to the fidelity obtained with Taq DNA Polymerase.
These characteristics make the Expand High FidelityPLUS PCR System blend the product of choice if a larger number of targets need to be amplified at the same time with high yield in combination with improved accuracy. In addition, the product can be used for the efficient labeling of DNA fragments with radioactive or non-radioactive modified nucleotides.

Application

  • The Expand High FidelityPLUS PCR System is designed for the robust amplification of fragments up to 5 kb from all types of DNA with outstanding yield, specificity, sensitivity, and accuracy. This convenient dNTPack is the product of choice for the development of a new amplification assay or the optimization of an existing assay. PCR
  • RT-PCR
  • Primer extension
  • Carryover prevention
It has been used in the ultradeep pyrosequencing methods, inverse PCR and to measure the repeat length of CGG sequences.
Twofold higher yield and threefold greater fidelity are obtained compared to Taq DNA Polymerase. Use Expand High FidelityPLUS PCR System for:
  • PCR up to 5kb
  • RT-PCR up to 5kb

Features and Benefits

Expand High FidelityPLUS PCR System is a mixture of Taq DNA polymerase and a DNA polymerase with proofreading activity. The Expand High FidelityPLUS PCR System shows a twofold increase in fidelity compared to the Expand High Fidelity PCR System, and a sixfold increase compared to Taq DNA Polymerase.
By incorporating dUTP, Uracil-DNA Glycosylase can be used to prevent PCR product carryover. Use this kit for efficient labeling of DNA with radioactive or non-radioactive nucleotides. It is optimized for PCR of DNA fragments up to 5kb.
Use 2.5U for a standard 50μl PCR.

  • Safeguard against cross-contamination.
This is the choice for large numbers of amplifications.
  • Obtain sixfold higher fidelity and higher yields,
compared to Taq DNA Polymerase.
  • Efficiently label DNA.
Use this kit to label DNA with radioactive or nonradioactive nucleotides.
  • Cost-effective.
Use the convenient solution of premixed PCR grade dNTPs.

The HIV-1 nucleoside RT inhibitor (NRTI)-resistance mutation, K65R confers intermediate to high-level resistance to the NRTIs abacavir, didanosine, emtricitabine, lamivudine, and tenofovir; and low-level resistance to stavudine. Several lines of evidence suggest that K65R is more common in HIV-1 subtype
