General description
The Dione MicroMap Kit provides a method for small molecule intracellular target identification via photocatalytic proximity labeling. The kit contains a catalyst that can be coupled to an acid on almost any small-molecule ligand. The subsequent ligand-Iridium conjugate can be used to perform multiple target identification experiments. The following kit provides enough Ir-catalyst to synthesize 20 μMoles of iridium–ligand conjugate and enough Diazirine-PEG3-Biotin to perform 10 proteomics experiments.
The kit provides a step-by-step procedure for conjugating your ligand of choice to a photocatalyst that localizes light-mediated crosslinking to the ligands target. The ligand–iridium conjugate can then be incubated with live cells along with a biotinylated diazirine reagent. Analogous to a satellite, the photocatalyst localizes carbene generation from the diazirine only in the direct vicinity and thus covalently modifies the target protein in a catalytic manner, amplifying the signal for downstream analysis.
To link to an amine on a small molecule (instead of an acid), the Hyas Kit is available.
