Clone 2K14-2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Coilin (COIL). It targets an epitope within 25 amino acids from the C-terminal region.

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human Coilin (COIL).

Anwendung

Quality Control Testing

Evaluated by Western Blotting with His-ABP-tagged recombinant fragment of Human Coilin (COIL).

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected His-ABP tagged recombinant fragment of Human Coilin (COIL).

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Coilin (COIL) in HepG2 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected Coilin (COIL) in Human testis tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound Coilin peptide with a KD of 3.0 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected COIL in HepG2 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Zielbeschreibung

Coilin (UniProt: P38432; also known as p80-coilin) is encoded by the COIL (also known as CLN80) gene (Gene ID: 8161) in human. Coilin is a nuclear phosphoprotein that is express in all cell types. It is a component of nuclear coiled bodies (Cajal bodies) that are involved in the modification and assembly of nucleoplasmic snRNPs. It is shown to be hyperphosphorylated during mitosis and this hyperphosphorylation triggers Cajal bodies disassembly during cell replication. Sixteen different phosphorylated residues have been identified in coilin, five of these are found primarily in only mitotic cell preparations and eight in only interphase samples. Its first N-terminal 92 amino acids are essential for self-association leading to its characteristic fibrous pattern observed in Cajal bodies. In the central region it has two nuclear localization signals and a nucleolar localization sequence. The conserved C-terminal region contains an atypical Tudor domain (aa 460-559) that contains four characteristic β-strands (β1β2 and β3β4) and one short 310 helix. The final ten residues at the C-terminal region have been implicated in controlling availability of the N-terminus for self-interaction, Cajal bodies formation, and coilin localization. Symmetrical dimethylation of arginine residues within the RG (arginine-glycine) repeat region (aa 423-420) of coilin is reported to enhance its affinity for SMN, and thus localization of SMN complexes to Cajal bodies. Coilin is suggested to promote RNP biogenesis by acting as a chaperone of nuclear small non-coding RNAs, which might not be essential in differentiated cells, but may become critical in rapidly developing embryos. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstituierung

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
