Allgemeine Beschreibung

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Spezifität

Expected to react with most other vertebrate species based on highly conserved sequence homology.
This antibody only detects histone H3 when trimethylated on Lys4.

Immunogen

Epitope: Trimethylated Lys4 (H3K4me3).
Synthetic peptide corresponding to residues surrounding and including trimethlyated Lys4 of histone H3.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected detected histone H3 Lys4 trimethylation in HeLa cells.
The unconjugated antibody (Cat. No. 07-473) is shown to be suitable also for dot blot, chromatin immunoprecipitation (ChIP), ChIP-seq, and Western blotting applications.
This Anti-trimethyl Histone H3 (Lys4) Antibody, Alexa Fluor 555 Conjugate is validated for use in Immunocytochemistry for the detection of trimethyl Histone H3.

Qualität

Evaluated by Immunocytochemistry in HepG2 cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected histone H3 Lys4 trimethylation in HepG2 cells.
