Specificity

Clone 1G22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects CXCR5. It targets an epitope within 19 amino acids from the N-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal, extracellular domain of human CXCR5.

Application

Quality Control Testing

Evaluated by Immunocytochemistry in Raji cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected CXCR5 in Raji cells.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected CXCR5 in one million Human peripheral blood mononuclear cells (PBMC) co-stained with CD19APC .

Affinity Binding Assay: A representative lot of this antibody bound CXCR5 peptide with a KD of 1.5 x 10-7 in an affinity binding assay.

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CXCR5 in HL-60 cell lysate.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

C-X-C chemokine receptor type 5 (UniProt: P32302; also known as CXC-R5, CXCR-5, Burkitt lymphoma receptor 1, Monocyte-derived receptor 15, MDR-15, CD185) is encoded by the CXCR5 (also known as BLR1, MDR15) gene (Gene ID: 643) in human. CXCR5 is a multi-pass membrane protein of the G-protein coupled receptor 1 family with seven transmembrane domains. Its expression is observed in mature B cells, Burkitt lymphoma cells, and in T-follicular helper T cells (TFH). It serves as a receptor for CXCL13, the chemokine that is essential for secondary lymphoid tissue orchestration and lymphoid neogenesis. Both CXCL13 and CXCR5 are essential for trafficking B cells and homing B cells to lymphoid tissues and for the embryonic development of the majority of lymph nodes and Peyer′s patches. Transgenic mice deficient in CXCL13 or CXCR5 exhibit severely impaired lymph node development, lacking peritoneal B lymphocytes, and deficient in circulating antibodies to common bacterial antigens. CXCR5 expression is also observed in retinal pigment epithelial cells (RPE) and is required for maintaining retinal pigment epithelial (RPE) cells homeostasis and blood-retinal barrier through PI3K-AKT-FOXO1 pathway. Two isoforms of CXCR5 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
