General description

Cadherin 5 (CDH5), also known as cadherin 144 (CD144), is encoded by the gene mapped to long arm of human chromosome 16. The encoded protein is expressed in endothelial cells, but it absent in normal epithelium.
VE-Cadherin (also designated vascular endothelial cadherin, cadherin-5, and cluster of differentiation 144 (CD144)) is a non-type I or II cadherin that is expressed on endothelial cells of all blood and lymphatic vessels, where it organizes intercellular adherens junctions.

Biochem/physiol Actions

Cadherin 5 (CDH5)/CD144 plays an essential role in various cellular processes, such as regulation of cell adhesion, cell proliferation, cell survival, cell shape, cell motility. Additionally, it is also involved in the regulation of signaling pathways and transcriptional gene regulation. CD144/VE-cadherin is a core component of endothelial adherens junction, which plays a vital role in angiogenic sprouting by enhancing cell migration and cell elongation. CD144 expressed in glioblastoma multiforme (GBM) facilitates the transdifferentiation of glioblastoma stem like cells (GSCs) to endothelial cells and form blood vessels, under hypoxia condition. VE-cadherin induces endothelial cell-cell adhesion at adherens junctions (AJs). CD144 gene knockout results in loss of cell polarity and modification in vascular lumenal structure. CD144 inhibits the synthesis of CDH2 (also known as N-cadherin) in the endothelium.
VE-cadherin (CD144) interacts with β-catenin, plakoglobinand Adherens junction protein (p120ctn).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
