General description

apo-Transferrin (aTF) human can be supplemented with iron or used to bind free iron present in media. It can be used to transport iron into mammalian cells.

Application

apo-Transferrin human has been used as a supplement:
  • in Dulbecco′s modified Eagle medium/HamF12 (DMEM/F-12) to culture human adrenocortical carcinoma cells
  • in DMEM/F-12 to culture neural precursor cells from mice
  • in collecting medium NCSU23 to culture preantral follicles from porcine ovaries

Biochem/physiol Actions

apo-Transferrin (aTF) is an endogenous iron chelator and shows protective effects in the host by scavenging free iron and preventing the formation of toxic reactive oxygen species (ROS). It shows a therapeutic effect against thalassemia. After the release of iron, receptor-bound apo-transferrin reaches the cell surface instead of being transported to lysosomes for degradation. The cell surface has a neutral pH that promotes the release of apo-transferrin from its receptor, where it can again circulate and bind additional iron to undergo further rounds of iron delivery to cells.

Other Notes

Non-heme iron-transport protein.
