General description

HGS (hepatocyte growth factor-regulated tyrosine kinase substrate) protein has a molar mass of 110kDa and contains a double zinc-finger motif. It is localized in the cytoplasm. It is a 777-amino-acid protein with 93% homology to mouse Hrs. The gene is mapped to human chromosome 17q25.

Application

Biochem/physiol Actions

HGS (hepatocyte growth factor-regulated tyrosine kinase substrate) gene encodes a FYVE domain protein localized to early endosomes. It functions in membrane trafficking of various molecules. It recruits Smad2 and Smad3 to the activin receptor complex in cooperation with SARA and regulates Smad signaling involved in mouse early embryonic development. It functions in the regulation of endosomal sorting and targets proteins for lysosome-dependent degradation. Rapid phosphorylation of Hgs is stimulated by IL-2, granulocyte-macrophage colony-stimulating factor and hepatocyte growth factor. It is associated with STAM, a signal-transducing adaptor molecule, and suppresses its function in cell growth signaling mediated by the cytokines.
