General description

Inosine-5′-monophosphate dehydrogenase 1 (IMPDH 1) catalyzes the conversion of inosine 5′-phosphate (IMP) to xanthosine 5′-phosphate (XMP), as part of the de novo synthesis of guanine nucleotides. It therefore is crucial in the regulation of cell growth. IMPDH 1 may also have single-stranded nucleic acid-binding activity involving it in RNA and/or DNA metabolism, and is suspected to play a role in the growth progression of some tumors. Defects in IMPDH 1 abnormalities are linked to retinitis pigmentosa type 10 (RP10) and Leber congenital amaurosis type 11 (LCA11). Due to the connection between IMPDH activity and cell proliferation, it has been become a target for cancer and viral chemotherapy and immunosuppressive drugs. The activities of the antitumor drug tiazofurin, the antiviral drug ribavirin, and the immunosuppressive drugs mizoribine and mycophenolic acid (MPA) are attributed to the inhibition of IMPDH. In addition, bacterial and parasitic IMPDH′s differ significantly from mammalian enzymes, which make it a suitable target for anti-infective drugs.
