General description

Lysine-specific demethylase 5C (UniProt P41229; also known as Histone demethylase JARID1C, Jumonji/ARID domain-containing protein 1C, Protein SmcX, Protein Xe169) is encoded by the KDM5C (also known as DXS1272E, JARID1C, MRXSCJ, SMCX, XE169) gene (Gene ID 8242) in human. RBP2 (JARID1A), PLU-1 (JARID1B), KDM5C (JARID1C), and SMCY (JARID1D) constitute the Jumonji/ARID (JARID) subfamily of JmjC-containing proteins. KDM5C/JARID1C catalyzes the demethylation of di- and trimethylated lysine 4 of histone 3 in a Fe(II) and α-ketoglutarate-dependent manner. JARID1 catalytic activity is medidated by its JmjC domain (a.a. 468-634), while the JmjN domain (a.a. 14-55) and the C5HC2 zinc finger (a.a. 326-372 & 1187-1248) are both important in assisting JARID1C catalytic activity. JARID1C also contains an AT-rich domain-interacting domain (ARID) that binds DNA and its N-terminal PHD (plant homeodomain) finger (a.a. 326-372) is reported to bind trimethylated lysine 9 on histone 3 (H3K9me3). JARID1C interacts with PCNA through its PCNA-interaction protein (PIP) motif and JARID1C is among 485 chromatin factors enriched at nascent versus mature post-replicative chromatin, indicating a potential transcription-independent role during DNA replication.

Specificity

Clone 2B4.2 targets an N-terminal region sequence present in all human and murine JARID1C/KDM5C spliced isoforms reported by UniProt (P41229 and P41230).

KLH-conjugated linear peptide corresponding to an N-terminal region sequence of human JARID1C.

This Anti-Histone Demethylase JARID1C Antibody, clone 2B4.2 is validated for use in Western Blotting for the detection of Histone Demethylase JARID1C.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 2.0 µg/mL of this antibody detected histone demethylase JARID1C in 10 µg of HeLa cell lysate.

Target description

~180 kDa observed. 175.7/175.3 kDa (human/mouse isoform 1), 175.4/175.0 kDa (human/mouse isoform 2), 170.6/170.2 kDa (human/mouse isoform 3), 155.1/175.6 kDa (human isoform 4/5) calculated. Uncharacterized band(s) may appear in some lysates.
