General description

Dolichyl-diphosphooligosaccharide-protein glycosyltransferase subunit 1 (EC 2.4.99.18; UniProt P04843; also known as Dolichyl-diphosphooligosaccharide-protein glycosyltransferase 67 kDa subunit, Ribophorin I, RPN-I, Ribophorin-1) is encoded by the RPN1 gene (Gene ID 6184) in human. Ribophorin I is an ER transmembrane protein and a subunit of the mammalian Oligosaccharyltransferase (OST) complex. Ribophorin I binds newly synthesized target proteins and delivers them to the catalytic core of the OST complex. Association of ribophorin I with newly synthesized membrane proteins was observed after the exit of the nascent chain from the Sec61 translocon, and it was unaffected by the N-glycosylation status. Ribophorin I is reported to preferentially interact with misfolded over native ribonuclease A. In addition, Ribophorin I is shown to complex with another ER protein malectin and enhance malectin′s ability to prevent the secretion of the folding-defective proteins.

This Anti-Ribophorin I/RPN-I Antibody is validated for use in Western Blotting for the detection of Ribophorin I.
Western Blotting Analysis: An 1:5,000 dilution from a representative lot detected ribophorin I in in 25 µg of HeLa cell lysate (Courtesy of Dr. Christopher V. Nicchitta, Duke University Medical Center, Durham, NC).
Western Blotting Analysis: A representative lot detected ribophorin I in both the smooth and rough microsomes sub-fractionated from a rat liver microsome preparation (Menon, A.K., et al. (2000). Curr. Biol. 10(5):241-252).
Western Blotting Analysis: A representative lot detected ribophorin I in canine rough microsome (RM) fractions obtained by sucrose gradient centrifugation of digitonin- or DHPC-solubilized canine RM preparation with or without ribosomes stripping by EDTA treatment (Potter, M.D., and Nicchitta, C.V. (2000). J. Biol. Chem. 275(3):2037-2045).

Quality

Evaluated by Western Blotting in canine pancreas tissue rough microsome lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected Ribophorin I/RPN-I in 2.5 µL of canine pancreas tissue rough microsome lysate.
