Description générale

Clone 1J17 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TRIM14. It targets an epitope within 21 amino acids from the N-terminal half.

Evaluated by Western Blotting in HeLa cell lysate.



Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected TRIM14 in HeLa cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TRIM14 in human liver tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound TRIM14 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected TRIM14 in HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Tripartite motif-containing protein 14 (UniProt: Q14142) is encoded by the TRIM14 (also known as KIAA0129) gene (Gene ID: 9830) in human. The protein encoded by TRIM14 is a member of the tripartite motif (TRIM) family. It plays an essential role in the innate immune defense against viruses and bacteria. TRIM14 includes zinc-binding domain (B-box type) and two coiled-coil regions. The B-box zinc finger is responsible for inhibition of SPI1-mediated transcriptional activation. It lacks the N-terminal RING domain, which is found in most other proteins in the TRIM family. TRIM14 expressed in various tissues with highest expression in liver and also in immune-related tissues such as the spleen, thymus, and peripheral blood leukocytes and is undetectable in skeletal muscle. The protein localizes to cytoplasmic bodies; mitochondria and cytoplasmic vesicle (phagosomes). Four alternatively spliced transcript variants for this gene have been described. It is involved in various cellular processes and key functions include Antiviral defense, Autophagy, Inflammatory Response and Cell Proliferation and Survival. TRIM14 is crucial in the host defense against viral infections. Its antiviral activity is significant in the context of infections by viruses such as HIV, influenza, and hepatitis B and C. The antiviral mechanism is by IFN-stimulated genes and Cytokine Signaling in Immune system. TRIM14 is also known to be associated with Spondyloepimetaphyseal Dysplasia of Geneviève Type. TRIM14 may contribute to the pathogenesis of autoimmune diseases if its regulatory mechanisms are disrupted. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
