Spécificité

Clone 2I8 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Retinoid X receptor alpha. It targets an epitope within 16 amino acids from the N-terminal region.

Immunogène

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal region of human retinoid X receptor alpha.

Application

Anti-RXR-alpha, clone 2I8 ZooMAb, Cat. No. ZRB1519, is a recombinant Rabbit monoclonal antibody that targets RXR-alpha and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RXR-alpha in MCF-7 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RXR-alpha in HUVEC and HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected RXR-alpha in human skin and human esophagus tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound RXR alpha peptide with a KD of 1.3 x 10-6 in an affinity binding assay.

Description de la cible

Retinoic acid receptor RXR-alpha (UniProt: P19793; also known as Nuclear receptor subfamily 2 group B member 1, Retinoid X receptor alpha) is encoded by the RXRA (also known as NR2B1) gene (Gene ID: 6256) in human. RXR-alpha is a homodimeric protein that can also for heterodimer with RAR-alpha via its C-terminal. RXR-alpha serves as a receptor for retinoic acid and binds to target response elements in response to their ligands, all-trans or 9-cis retinoic acid, to regulate the expression of genes involved in various biological processes. 9-cis retinoic acid is considered as a high affinity ligand of RXR-alpha. The RAR/RXR heterodimers are reported to bind to the retinoic acid response elements (RARE) composed of tandem 5′-AGGTCA-3′ sites known as DR1-DR5 to regulate transcription. In the absence of ligand, the RXR-RAR heterodimers associate with a multiprotein complex containing transcription co-repressors that induce histone deacetylation, chromatin condensation and transcriptional suppression. On ligand binding, the corepressors dissociate from the receptors and coactivators are recruited leading to transcriptional activation. RXR-alpha is highly expressed in liver and its expression has also been reported in lung fibroblasts, monocytes, kidney, and brain. RXR-alpha is composed of three domains: a modulating N-terminal domain (AF1 domain), a DNA-binding domain, and a C-terminal ligand-binding domain (AF2 domain). Its DNA binding region is localized to amino acids 135-200. It has two NR-C4 type zinc-finger domains (aa 135-155 and 171-195). Its nuclear localization signal is localized to amino acids 160-165 and the region comprising of amino acids 348-367 is essential for its nuclear export. Its nuclear localization is enhanced by vitamin D3. Its acetylation by EP300 is shown to enhance its DNA-binding and transcriptional activity. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
