Specificity

Clone 20A11.1 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Werner′s syndrome protein (WRN).

Immunogen

GST-tagged recombinant protein corresponding to human Werner syndrome protein (WRN).

Application

Quality Control Testing

Evaluated by Western Blotting in Ramos cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected WRN in Ramos cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected WRN in HEK293 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected WRN in MOLT-4 cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected WRN in one million Ramos cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Bifunctional 3′-5′ exonuclease/ATP-dependent helicase WRN (UniProt: Q14191; also known as DNA helicase RecQ-like type 3, RecQ protein-like 2, Werner syndrome protein) is encoded by the WRN (also known as RECQ3, RECQL2) gene (Gene ID: 7486) in human. WRN is a multifunctional, monomeric, enzyme that exhibits both magnesium and ATP-dependent DNA-helicase activity and 3′->5′ exonuclease activity towards double-stranded DNA with a 5′-overhang. It can also form homooligomers. It plays a role in repair of double-strand DNA following -irradiation. It does not have any nuclease activity towards single-stranded DNA or blunt-ended double-stranded DNA. It binds preferentially to DNA substrates containing alternate secondary structures, such as replication forks and Holliday junctions. It may also play a role in the dissociation of joint DNA molecules that can arise as products of homologous recombination, at stalled replication forks or during DNA repair and alleviates stalling of DNA polymerases at the site of DNA lesions. Mutations in WRN gene have been linked to development of colorectal cancers and Werner syndrome that is characterized by the premature onset of multiple age-related disorders. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
