General description

Glycolysis is one of the major metabolic pathways cells undergo to produce energy and results in the production of pyruvate. One of the eventual end products of this process is lactate dehydrogenase-mediated conversion of pyruvate to L-lactate via lactic acid fermentation. This enzymatic mechanism allows L-lactate to serve as an indicator of glycolysis.

Application

The Glycolysis Assay Kit may be used for:
  • Cancer Research
  • Diabetes Research
  • Cardiovascular Disease Research

Features and Benefits

Broad Detection Range: Detect L-lactate levels accurately up to 10 mM, using only 5 μL of sample in a 96-well plate setup, ensuring comprehensive analysis with minimal sample requirement.

Simplified Process: Experience a streamlined process with the addition of only a single working reagent and a room temperature reaction, reducing complexity and saving valuable time and effort.

Compatibility with High-Throughput Systems: Easily incorporate our kit into high-throughput handling systems, ensuring smooth and accurate processing, enhancing efficiency in your laboratory workflow.

Suitability

The kit is suitable for the direct determination of L-Lactate produced by glycolysis in cell media samples and for the screening of glycolysis inhibitors.

Principle

The Glycolysis Assay Kit is based on measuring the production of L-Lactate from glycolysis in cells. L-Lactate that is secreted into the cell media is quantified using a coupled reaction involving the lactate dehydrogenase catalyzed oxidation of L-lactate that generates pyruvate and NADH, which subsequently reduces a formazan dye. The intensity of the reduced dye, measured at 565 nm, is directly proportional to the L-lactate concentration in the sample, which in turn is directly proportional to the glycolytic rate of the cells.
