General description

Rho guanine nucleotide exchange factor 9 (UniProt O43307; also known as Collybistin, PEM-2 homolog, Rac/Cdc42 guanine nucleotide exchange factor 9) is encoded by the ARHGEF9 (also known as ARHDH9, EIEE8, KIAA0424) gene (Gene ID 23229) in human. Guanine nucleotide exchange factors (GEFs) are cytoplasmic proteins that activate GTPases by stimulating the release of guanosine diphosphate (GDP) to allow binding of guanosine triphosphate (GTP). ARHGEF9 acts as a specific GEF for CDC42 and promotes formation of GPHN clusters. In the brain tissue it interacts with the receptor-anchoring protein gephyrin to activate dc42, which is known to regulate actin cytoskeleton dynamics. Its binding to gephyrin can also modulate GABAergic and glycinergic neurotransmission at the synapse. Abnormalities in its expression have been linked to epileptic encephalopathy, early infantile, 8.

Specificity

Clone 4D7.1 targets ARHGEF9 C-terminal region present in human isoforms 1/2/3 and mouse isoforms 1/2/4 reported by UniProt (O43307 and Q3UTH8). Reactivity toward mouse isoform 3 is not expected.

Target description

~61 kDa observed. 60.98/54.97/49.46 kDa (human isoform 1/2/3) and 60.93/58.62/60.86 kDa (mouse isoform 1/2/4) calculated. Human isoforms 1 and 2 are also known as CB3(SH3+) and CB3(SH3-). Uncharacterized bands may be observed in some lysate(s).
