General description

Helicase-like transcription factor (UniProt Q14527; also known as DNA-binding protein/plasminogen activator inhibitor 1 regulator, HIP116, RING finger protein 80, Sucrose non-fermenting protein 2-like 3, SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A member 3) is encoded by the HLTF (also known as HIP116A, RNF80, SMARCA3, SNF2L3, ZBU1) gene (Gene ID 6596) in human. Helicase-like transcription factor (115 kDa) is a SWI/SNF (mating-type switching/sucrose non-fermenting) protein that uses ATP hydrolysis to remodel chromatin in multiple cellular processes. It is shown to regulate transcription by binding to the SV40 enhancer and the promoters Serpin E1 and HIV-1. It is well expressed in brain, heart, kidney, liver, lung, pancreas, placenta, and skeletal muscle. It was initially characterized for its DNA binding and transcriptional activity, however, later was shown to act as an E3 ubiquitin ligase and was implicated in post-replication DNA repair by polyubiquitination of proliferating cell nuclear antigen (PCNA). The inactivation of HIP116 is shown to induce an elevated level of chromosome breaks and fusions in mouse embryonic fibroblasts after treatment with alkylating agents. HIP116 inactivation in transgenic mice is reported to disrupt cell cycle at the G2/M phase.

Specificity

Clone 11-1 detected predominantly nuclear HLTF immunoreactivity by immunohistochemistry staining. Clone 11-1 epitope lies within the N-terminal region present in human HLTF (SMARCA3) spliced isoform 1, but not in isoform 2 reported by UniProt (Q14527).
