Spezifität

Clone 3N8 is a ZooMAb® rabbit recombinant monoclonal antibody that detects SIRPA. It targets an epitope within 26 amino acids from the C-terminal extracellular domain. Might crossreact with SIRPB1-Isoform 3.

KLH-conjugated linear peptide corresponding to 26 amino acids from the C-terminal extracellular domain of human SIRPA.

Anwendung

Quality Control Testing

Evaluated by Western Blotting in U-937 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SIRPA in U-937 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SIRPA in SK-MEL-28 and human PBMC lysates.

Affinity Binding Assay: A representative lot of this antibody bound SIRPA peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SIRPA in U-937 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SIRPA in human cerebral cortex tissue sections.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected SIRPA in one million human PBMCs.

Tyrosine-protein phosphatase non-receptor type substrate 1 (UniProt: P78324; also known as SHP substrate 1, SHPS-1, Brain Ig-like molecule with tyrosine-based activation motifs (Bit), CD172 antigen-like family member A, Inhibitory receptor SHPS-1, Macrophage fusion receptor, MyD-1 antigen, Signal-regulatory protein alpha-1 (Sirp-alpha-1), Signal-regulatory protein alpha-2 (Sirp-alpha-2), Signal-regulatory protein alpha-3 (Sirp-alpha-3), p84) is encoded by the SIRPA (also known as BIT, MFR, MYD1, PTPNS1, SHPS1, SIRP) gene (Gene ID: 140885) in human. Tyrosine-protein phosphatase non-receptor type substrate 1 (SIRPA) is a type I membrane protein and is characterized by its immunoglobulin-like domains located in the extracellular region, a single transmembrane segment, and a cytoplasmic region containing immunoreceptor tyrosine-based inhibition motifs (ITIMs). SIRPA is primarily involved in negative regulation of immune cell activation. It acts as a receptor for CD47, a signal-regulatory protein expressed on the surface of many cells. When engaged with CD47, SIRPA transmits a signal that inhibits phagocytosis by macrophages, thus playing a crucial role in self-recognition and protection of healthy cells from immune clearance. Additionally, the ITIM domains of SIRPA recruit and activate phosphatases such as SHP-1 and SHP-2, which then modulate downstream signaling pathways, including those involved in the inhibition of cellular activation and proliferation. SIRPA is widely expressed on cells in the immune system, including macrophages, dendritic cells, neurons, and other tissue cells. Alterations in SIRPA signaling are implicated in a variety of diseases. In cancer, cells often overexpress CD47 to engage SIRPA on immune cells, thereby evading phagocytosis and promoting tumor survival and growth. Furthermore, dysregulation of SIRPA has been associated with autoimmune diseases, where impaired signaling may contribute to the destruction of healthy cells.

Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
