Spezifität

Clone 2N22 is a Rabbit recombinant monoclonal antibody that specifically detects IKKb. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human IKKb.

Anwendung

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IKKβ in A549 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IKKβ in HepG2, L6, and NIH3T3 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound IKKb with a KD of 7.9 x 10-8 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Various dilutions of this antibody detected IKKβ in ELISA application.

Zielbeschreibung

Inhibitor of nuclear factor kappa-B kinase subunit beta (UniProt: O14920; also known as EC:2.7.11.10, I-kappa-B-kinase beta, IKK-B, IKK-beta, IkBKB, I-kappa-B kinase 2, IKK2, Nuclear factor NF-kappa-B inhibitor kinase beta, NFKBIKB, Serine/threonine protein kinase IKBKB) is encoded by the IKBKB (also known as IKKB) gene (Gene ID: 3551) in human. The IKK complex is composed of three subunits, IKKa , IKKb, and IKKg (NEMO). While IKKa and IKKb are highly similar catalytic subunits and can phosphorylate IkB, IKKg is the regulatory subunit. IKKb is a serine kinase that plays an essential role in the NF-kB signaling pathway. It acts as a part of the canonical IKK complex in the conventional pathway of NF-kB activation. Its protein kinase domain is localized in amino acids 15-300. It phosphorylates inhibitors of NF-kB (IkB) on two critical serine residues that allow polyubiquitination of these inhibitors and their subsequent degradation by the proteasome. This allows free NF-kB to translocate into the nucleus and activate the transcription of genes involved in immune response, growth control, and protection against apoptosis. Upon cytokine stimulation, IKKb is phosphorylated on serine 177 and serine 181 by MEKK1 and/or MAP3K14/NIK as well as TBK1 and protein kinase Cz that enhances its activity. Once activated, it undergoes autophosphorylation on the C-terminal serine cluster, which reduces its activity and prevents prolonged activation of the inflammatory response. Mutations in IKBKB gene are known to cause life threatening immunodeficiency that develops in in infancy and increases susceptibility to bacterial, fungal, and viral infections. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. (Ref.: Li, ZW., et al. (1999). J. Exp. Med. 189(11); 1839-1845).

Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Rekonstituierung

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
