Spezifität

Clone 3F22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Siglec-9. It targets an epitope within 15 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal, cytoplasmic domain of human Siglec-9.

Anwendung

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Siglec-9 in Jurkat cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Siglec-9 in HepG2 and Raw264.7 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Siglec-9 in human spleen tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected Siglec-9 in one million Human peripheral blood monouclear cells (PBMC).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
Anti-Siglec-9, clone 3F22 ZooMAb®, Cat. No. ZRB1031, is a recombinant Rabbit monoclonal antibody that detects Siglec-9 and is tested for se in Flow Cytometry, Immunohistochemistry (Paraffin), and Western Blotting.

Zielbeschreibung

Sialic acid-binding Ig-like lectin 9 (UniProt: Q9Y336; also known as Siglec-9, CDw329, Protein FOAP-9, CD329) is encoded by the SIGLEC9 (also known as UNQ668/PRO1302) gene (Gene ID: 27180) in human. Sialic-acid-binding immunoglobulin-like lectins (Siglecs) belong to a lectin family of surface receptors that are predominantly expressed on hematopoietic cells with cell type and differentiation specificity. By binding with sialic acids at the terminal position of glycoproteins and lipids they regulate the intensity and duration of innate or adaptive immune response. Siglec-9 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-17), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 18-348), a transmembrane domain (aa 349-369), and a cytoplasmic domain (aa 370-463). It has one Ig-like V-type domain (aa 20-140) and two Ig-like C2-type domains (aa 146-229 and 236-336). It also contains one ITIM motif (aa 431-436) that is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-continaing phosphatases. It is expressed by peripheral blood leukocytes and is also found in a variety of other tissues at lower levels. Siglec9 serves as a putative adhesion molecule that mediate sialic-acid dependent binding to cells. It preferentially binds to alpha-2,3- or alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. Reduced expression of Siglec-9 on natural killer cells has been linked to persistent hepatitis B virus replication. Increased expression of soluble Siglec-9 has been reported in subjects with chronic obstructive pulmonary disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Rekonstituierung

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
