Allgemeine Beschreibung

PU.1 is a transcription factor of the Ets family and activates target genes through the purine-rich PU-box. PU.1 plays an important role for the differentiation of myeloid cells and early B cell lymphocytes. PU.1 is overexpressed in some B cell lymphoma, Hodgkin, and non- Hodgkin lymphoma.
Spi-1 proto-oncogene (SPI1), also known as PU.1, is a mammalian transcription factor. It is encoded by the gene mapped to human chromosome 11p11.2. The encoded protein belongs to the E26 transformation specific (Ets) family of transcription factors. SPI1 is characterized with a C-terminal domain, an N-terminal transactivation domain and a central PEST domain involved in protein-protein interaction. The protein is mainly expressed in cells of the hematopoietic lineage such as macrophages, granulocytes and B lymphocytes.

Spi-1 proto-oncogene (SPI1)/ PU.1 plays an essential role in myeloid development. It also acts as a transcription activator and is involved in macrophage proliferation. In addition, PU.1 along with other transcription factors, plays a vital role in fetal hematopoiesis. PU.1 controls cellular communication in the immune system. Thus, loss/reduced gene expression leads to loss of cellular communication, which in turn is associated with the development of leukemia.
