Allgemeine Beschreibung

The protein Death-associated protein 1 or also known as DAP-1/DAP1 and encoded by the gene DAP/DAP1 is a negative regulator of autophagy. The link of DAP1 to autophagy was recognized because in DAP1 knockdowns there was enhanced autophagic flux and DAP1 displayed a rapid decline in its phosphorylation in response to amino acid starvation, a key inducer of autophagy. Mapping of the phosphorylation sites and analysis of phosphorylation mutants indicated that DAP1 is functionally silenced in growing cells through mTOR-dependent phosphorylation on Ser3 and Ser51. Inactivation of mTOR during starvation caused a rapid reduction in these phosphorylation sites and converted the protein into an active suppressor of autophagy. These results are consistent with a "Gas and Brake" model in which mTOR inhibition also controls a buffering mechanism that counterbalances the autophagic flux and prevents its over activation under nutrient deprivation. DAP1 is also involved in mediating interferon gamma induced cell death, which appears to often be mediated by autophagy, and recently it has been shown to be important in regulating myogenesis and the apoptosis of satellite cells during development. DAP1/Death-associated protein 1 is regulated by phosphorylation; phosphorylation by MTOR inhibits the suppressive activity of DAP toward autophagy. DAP1 is expressed by most cells and up regulated in many cancer cells as well.
