Spécificité

Clone 1I16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Pancreas/duodenum homeobox protein 1 (PDX1). It targets an epitope within 17 amino acids from the N-terminal region.

Immunogène

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal region of human Pancreas/duodenum homeobox protein 1 (PDX1).

Application

Quality Control Testing

Evaluated by Western Blotting in Caco-2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PDX1 in Caco-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PDX1 in PANC-1 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PDX1 in PANC-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PDX1 in human pancreas tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound PDX1 peptide with a KD of 2.9 x 10-9 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Pancreas/duodenum homeobox protein 1 (UniProt: P52945; also known as PDX-1, Glucose-sensitive factor, GSF, Insulin promoter factor 1, IPF-1, Insulin upstream factor 1, IUF-1, Islet/duodenum homeobox-1, IDX-1, Somatostatin-transactivating factor 1, STF-1) is encoded by the PDX1 (also known as IPF1, STF1) gene (Gene ID: 3651) in human. PDX1 is a transcription factor that serves as a master regulator of pancreas organogenesis, maturation, and identity preservation of β-cells. It is involved in activation of insulin, somatostatin, glucokinase, and GLUT2 gene transcription. Although it is mainly expressed in β-cells and some -cells of the islets of Langerhans, its expression is also observed in duodenum. It is expressed early in pancreas development and persists through β-cell maturation. It is detected both in the nucleus and cytoplasm. Its nuclear localization signal resides in amino acids 197-203 and its homeobox domain is localized in amino acids 146-205. Glucose induces its nuclear translocation by triggering its phosphorylation via the PI3-kinase pathway. It is reported to be essential for compensatory β cell mass expansion in response to diet-induced insulin resistance through its roles in promoting β cell survival and compensatory hypertrophy. Mutations in PDX1 gene are linked to pancreatic agenesis that can lead to early-onset insulin-dependent diabetes mellitus (IDDM), as well as maturity onset diabetes of the young type 4 (MODY4). PDX1 deficient β cells show evidence of endoplasmic reticulum (ER) stress. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
