Clone 1L3 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Protein argonaute-1 (Ago 1). It targets an epitope within 21 amino acids from the internal region.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Ago 1 in NIH3T3 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Ago 1 in K562 and COS-7 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Ago 1 in human placenta tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound Ago1 peptide with a KD of 3.1 x 10-7 in an affinity binding assay.

Protein argonaute-1 (UniProt: Q9UL18; also known as Argonaute1, hAgo1, Argonaute RISC catalytic component 1, Eukaryotic translation initiation factor 2C 1, eIF-2C 1, eIF2C 1, Putative RNA-binding protein Q99) I encoded by the AGO1 (also known as EIF2C1) gene (Gene ID: 26523) in human. The highly conserved Argonaute proteins play a critical role in the regulation of gene expression and orchestrate the establishment and the maintenance of cell identity throughout the entire life cycle. In humans, four functional Argonaute proteins (Ago 1-4) have been described that exhibit high structure similarity, but also have a few non-conserved amino acids in their functional domains. In contrast to Ago 2, which has two N-terminal motifs that are required for its full catalytic activity, Ago1 contains only one N-terminal motif and Ago 3 and 4 lack these motifs and are catalytically inactive. About 80% of Ago1 clusters are associated with cell-type-specific transcriptional enhancers. Ago1 is required for RNA-mediated gene silencing. It binds to short RNAs such as miRNA or siRNA and represses the translation of mRNAs which are complementary to them. Its PIWI domain is localized in amino acids 515 to 816. Ago1 lacks RNA cleavage activity due to the absence of the conserved histidine at position 805. Ago1 is shown to be enriched around transcription start sites and binds preferentially to active transcriptional enhancers. Its association with enhancers is mediated by RNAs that are transcribed from these enhancers. However, the interaction of Ago1 with enhancers does not appear to regulate transcription of the neighboring genes. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
