Clone 1E18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Mitochondrial fission factor (MFF). It targets an epitope within 25 amino acids from the N-terminal half.

Immunogène

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human Mitochondrial fission factor (MFF).

Application

Quality Control Testing

Evaluated by Western Blotting in T47D cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MFF in T47D cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MFF in lysates from Human Brain and MOLT4 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MFF in human cerebral cortex tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound MFF peptide with a KD of 8.8 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MFF in C2C12 cells.

Description de la cible

Mitochondrial fission factor (UniProt: Q9GZY8; also known as MFF) is encoded by the MFF (also known as C2orf33, AD030, AD033, GL004) gene (Gene ID: 56947) in human. MFF is a single-pass type IV homodimeric membrane protein that has a cytoplasmic domain (aa 1-322), a helical domain (aa 323-340), and a mitochondrial intermembrane domain (aa 341-342). It is highly expressed in heart, kidney, liver, brain, muscle, and stomach. It is a homodimeric protein but may also interact with other proteins. It plays a role in mitochondrial and peroxisomal fission and promotes the recruitment and association of the fission mediator dynamin-related protein 1 (DNM1L) to the mitochondrial surface. It is also suggested to be involved in regulation of synaptic vesicle membrane dynamics by recruitment of DNM1L to clathrin-containing vesicles. MFF is shown to be an essential factor for mitochondrial recruitment of Drp1 during mitochondrial fission in mammalian cells and its down-regulation inhibits mitochondrial recruitment of Drp1 and induces mitochondria elongation. Exogenous expression of MFF shown to induce extensive mitochondrial fission. Abnormal activation of mitochondrial fission may drive the impairment of mitochondrial metabolic function that leads to inhibition of cancer stem cell propagation, and the activation of quiescence programs. Mutations in MFF gene can cause a form of encephalopathy that is characterized by delayed psychomotor development, severe hypotonia with inability to walk, and microcephaly. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
