Clone 1E9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Mitotic spindle assembly checkpoint protein MAD2A (MAD2). It targets an epitope within 18 amino acids from the C-terminal region.

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MAD2L1 in L6 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MAD2L1 in HeLa and A431 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MAD2L1 in human ovarian cancer tissue sections.

Affinity Binding Assay Analysis: A representative lot of this antibody bound Mad2L1 peptide with a KD of 1.8 x 10-7 in an affinity binding assay.

Description de la cible

Mitotic spindle assembly checkpoint protein MAD2A (UniProt: Q13257; also known as HsMAD2, Mitotic arrest deficient 2-like protein 1, MAD2-like protein 1, MAD2) is encoded by the MAD2L1 (also known as MAD2) gene (Gene ID: 4085) in human. MAD2L1 is a component of the mitotic spindle assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. It exists as a monomer or homodimer. It is reported to exist either in an open and inactive (O-MAD2) conformation or in the closed, active (C-MAD2) conformation. In the closed conformation it forms a heterotetrameric complex with MAD1L1 at unattached kinetochores during prometaphase, the complex recruits open conformation molecules of MAD2L1 (O-MAD2) and then promotes the conversion of O-MAD2 to C-MAD2. MAD2L1 undergoes phosphorylation on multiple serine residues by NIMA related kinase 2 (NEK2). The level of phosphorylation varies during the cell cycle and is shown to be the highest during mitosis. Phosphorylation abolishes interaction with MAD1L1 and reduces interaction with CDC20. Two isoforms of MAD2L1 are described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
