Description générale

Research area: Cell structure. Membrane Scaffold Protein 1D1 (MSP1D1) is a variant of MSP1 where the first eleven amino acids in the helix 1 portion are deleted of the original MSP1 sequence. It consists of an N-terminal histidine-tagged protein with a tobacco etch virus (TEV) protease cleavage site that is present between the protein sequence and histidine-tag. MSP1D1(–)variant of MSP1D1 lacks the histidine-tag and the TEV protease-recognition sequence.

Actions biochimiques/physiologiques

Nano-disctechnology is an approach that allows aqueous soluble membrane proteins in anative-like bilayer environment that enables the membrane proteins to remainactive and stable. The concept of this technology is derived from high-densitylipoprotein (HDL) particles and apolipoprotein, their primary proteincomponent. The nano-disc is a non-covalent structure consisting of MSP that isgenetically engineered to mimic the activity of apolipoprotein A1 (Apo-A1) andphospholipid bilayer. In the presence of synthetic phospholipids, MSP1D1 canself-assemble into discoidal nanoparticlesknown as nano-discs. These nano-discs are suitable for studying membraneproteins and are applicable in the field of medicine and biotechnology.
