Description générale

Tight junction protein 1/Zona occludens 1 protein (TJP1, ZO-1) is a transmembrane plaque protein belonging to the membrane-associated guanylate kinase (MAGUK) family of scaffold proteins. It consists of three PDZ (PSD-95/Discs large/ZO-1) domains, one SH3 (Src-homology 3) domain, and one GK (guanylate kinase) domain. Additionaly, it also contains a long proline-rich carboxyl terminal region and a ZU5 domain.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Actions biochimiques/physiologiques

Tight junction protein ZO-1 is a protein encoded by the TJP1 gene in humans and is mapped to chromosome 15q13. It is a membrane-scaffolding protein that plays an important role in maintaining tight-junction integrity, which is disrupted in many invasive cancers and intestinal diseases. It may act as a tumor-suppressor gene in GIST. It is an integral component of functional TJs and is regulated by p38δ. It shows a hypermethylation status in children with non-Hodgkin lymphoma (NHL) and the methylation level is positively correlated with the number of malignant cells in bone marrow. It may be useful as a novel molecular marker in early diagnosis, outcome assessment, prognostic evaluation and detection of minimal residual disease.
