Description générale

TNF-related apoptosis-inducing ligand (sTRAIL or Apo-2L) is the endogenous ligand for the ubiquitous pro-apoptotic TRAIL-R1 and TRAIL-R2 receptors. Upon ligand binding, TRAIL receptors form trimeric structures at the plasma membrane around a central zinc atom. The trimeric TRAIL receptors bind the FADD adaptor protein which in turn recruits caspase 8. TRAIL/APO-2L signals ultimately converge on caspase 3 and related proteins that promote cell death. Importantly, activated TRAIL receptors work cooperatively with death receptors 4 and 5 in transformed cells to induce caspase signaling, thereby preventing the proliferation of abnormal cells. TRAIL/ApO-2L has been implicated in pathophysiology of many disease processes, including cancer and autoimmune diseases.
