Description générale

Dolichyl-diphosphooligosaccharide-protein glycosyltransferase subunit 1 (EC 2.4.99.18; UniProt P04843; also known as Dolichyl-diphosphooligosaccharide-protein glycosyltransferase 67 kDa subunit, Ribophorin I, RPN-I, Ribophorin-1) is encoded by the RPN1 gene (Gene ID 6184) in human. Ribophorin I is an ER transmembrane protein and a subunit of the mammalian Oligosaccharyltransferase (OST) complex. Ribophorin I binds newly synthesized target proteins and delivers them to the catalytic core of the OST complex. Association of ribophorin I with newly synthesized membrane proteins was observed after the exit of the nascent chain from the Sec61 translocon, and it was unaffected by the N-glycosylation status. Ribophorin I is reported to preferentially interact with misfolded over native ribonuclease A. In addition, Ribophorin I is shown to complex with another ER protein malectin and enhance malectin′s ability to prevent the secretion of the folding-defective proteins.

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
