Specificity

Clone 1N21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Succinate receptor 1 (SUCNR1/GPR91). It targets an epitope within 18 amino acids from the third cytoplasmic domain in the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the third cytoplasmic domain within the C-terminal half of human Succinate receptor 1 (SUCNR1/GPR91).

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in Human breast cancer tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected SUCNR1/GRP91 in Human breast cancer tissue sections.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound SUCNR1/GPR91 peptide with a KD of 3.5 x 10^-8 in an affinity binding assay.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SUCNR1/GPR91 in human adipose tissue lysates.

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected SUCNR1/GPR91 in SUCNR1 and GST Proteins.

Immunofluorescence Analysis: A 1:5,000 dilution from a representative lot detected SUCNR1/GPR91 in Human liver and porcine kidney tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Succinate receptor 1 (UniProt: Q9BXA5; also known as G-protein coupled receptor 91, P2Y purinoceptor 1-like) is encoded by the SUCNR1 (also known as GPR91) gene (Gene ID: 56670) in human. Succinate receptor is a multi-pass membrane glycoprotein that belongs to the G-protein coupled receptor family with seven transmembrane domains. It serves as a receptor for succinate. SUCNR1 intracellular signaling is depended on the associated subunit type (G i, G s, and G q). Its expression is observed in kidney. Within the adaptive immune system, it has been detected in CD4+ and CD8+ T cells and in all subsets of B cells. T cells from patients with systemic lupus erythematosus (SLE) are reported to release large amounts of IL-10 and succinate following activation with oxidized mitochondrial DNA. Although its expression in the healthy CNS is reported to be low, its activation is considered as a key in the response of CNS cells to hypoxic-ischemic injury and inflammatory damage. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
