Specificity

Clone 1D6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human LMBR1L. It targets an epitope within 16 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal region of human LMBR1L.

Application

Quality Control Testing

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant Human LMBR1L.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound LMBR1L peptide with a KD of 5.8 x 10-8 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected recombinant Human LMBR1L.

Target description

Protein LMBR1L (UniProt: Q6UX01; also known as Limb region 1 protein homolog-like, Lipocalin-1-interacting membrane receptor, LIMR) is encoded by the LMBR1L (also known as KIAA1174, LIMR, UNQ458/PRO783) gene (Gene ID: 55716) in human. LIMBR1L is a multi-pass membrane protein with five extracellular domains, nine transmembrane domains, and five cytoplasmic domains. Its N-terminal region contains a lipocalin binding site (aa 1-76). It is shown to be highly specific for lipocalin 1 (Lcn1) and binds to Lcn1 with low micromolar to high nanomolar affinity and mediates its endocytosis. It is widely expressed in human tissues and plays an essential role in lymphocyte development by negatively regulating the canonical Wnt signaling pathway. It stabilizes the β-catenin destruction complex that is required for regulating catenin β1 levels. In murine models, the absence of LMBR1L is shown to cause aberrant activation of Wnt/β-catenin signaling, which leads to impaired development and functional defects in all lymphoid lineages. LMBR1L-deficient T cells are predisposed to apoptosis and die in response to antigen-specific expansion signals. Five isoforms of LMBR1L have been described that are produced by alternative splicing. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
