Specificity

Clone 1G2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects S100A4. It targets an epitope within 23 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the internal region of mouse S100A4.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected S100A4 in NIH3T3 cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected S100A4 in NIH3T3 cells.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected S100A4 in one million NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound S100A4 peptide with a KD of 5.4 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Protein S100-A4 (UniProt: P07091; also known as Metastasin, Metastatic cell protein, PEL98, Placental calcium-binding protein, Protein 18A2) is encoded by the S100a4 (also known as Capl, Mts1) gene (Gene ID: 20198) in murine species. The S100 protein family consists of 21 members that exhibit a high degree of sequence and structural similarity. These proteins are not functionally interchangeable and are involved in a wide range of biological processes. S100A4 is a homodimeric calcium-binding protein that plays a role in various cellular processes including motility, angiogenesis, cell differentiation, apoptosis, and autophagy. It increases cell motility and invasiveness by interacting with non-muscle myosin heavy chain IIA. It is reported to promote filament depolymerization and increases the amount of soluble myosin-IIA, resulting in the formation of stable protrusions facilitating chemotaxis. It is also reported to enhance the motility of macrophages, neutrophils, and leukocytes and promote the recruitment and chemotaxis of these cells to regulate inflammation and immune functions. S100A4 modulates the pro-apoptotic function of TP53 by binding to its C-terminal transactivation domain within the nucleus and reduce its levels. An increase in S100A4 expression has been correlated with a worse prognosis for patients with colorectal, gallbladder, bladder, esophageal, non-small-cell lung, gastric, medulloblastoma, pancreatic and hepatocellular cancers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
