Specificity

Clone 1B11 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects 78 kDa glucose-regulated protein (GRP-78). It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human 78 kDa glucose-regulated protein (GRP-78).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GRP-78 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected GRP-78 in lysates from U87-MG, RAW264.7, and NIH3T3 cells and recombinant full-length Human GRP-78 (without the signal peptide).

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected GRP-78 in human liver tissue sections.

Affinity Binding Assay Analysis: A representative lot of this antibody bound GRP-78 peptide with a KD of 9.8 x 10-6 in an affinity binding assay..

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Endoplasmic reticulum chaperone BiP (UniProt: P11021; also known as EC:3.6.4.10, 78 kDa glucose-regulated protein, GRP-78, Binding-immunoglobulin protein, BiP, Heat shock protein 70 family protein 5, HSP70 family protein 5, Heat shock protein family A member 5, Immunoglobulin heavy chain-binding protein) is encoded by the HSPA5 (also known as GRP78) gene (Gene ID: 3309) in human. GRP-78 is a multifunctional, abundant protein of the Hsp70 family. However, unlike the other members of this family, GRP-78 is not induced by heat shock because the promoter of GRP78 lacks the heat shock element. It is maintained at relatively low levels within the cell and its level increases significantly under stress condition that affect the endoplasmic reticulum (ER) and calcium homeostasis. It serves as an ER chaperone and exists either as a monomer or as a homooligomer. It plays a key role in protein folding and quality control in ER lumen. It is involved in the correct folding of proteins and degradation of misfolded proteins via its interaction with DNAJC10/ERdj5 and also acts as a key repressor of the ERN1/IRE1-mediated unfolded protein response (UPR). It is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that contains a nucleotide-binding domain (NBD; aa 125-280), a substrate-binding domain (aa 420-500), and a short interdomain linker (aa 409-419). The interdomain linker regulates its chaperone activity by mediating it homo-oligomerization. Its chaperone activity is regulated by ATP-induced allosteric coupling of the NBD and substrate-binding domains. GRP78 expression is shown to be up-regulated during early cardiogenesis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
