Gene Information

General description

Cyclin D1 (CCND1) is a proto-oncogene. It is one of the three D-type cyclins. The gene encoding it is localized on human chromosome 11q13. Cyclin dependent kinase-like 4 (CDKL4) is part of the cyclin-dependent protein kinase (CDK) family. The gene encoding it is localized on human chromosome 12q14-15.
Recombinant, full length human CDK4 (Genbank Accession No. NM_000075), amino acids 16-271, with C-terminal His-tag and modifications (amino acids 43G-47G were replaced with EE, and T172A), MW=34 kDa, and human Cyclin D1 (Genbank accession number NM_053056), MW 29 kDa, were co-expressed in a Baculovirus-infected Sf9 cells expression system.

Biochem/physiol Actions

Cyclin D1 (CCND1) regulates the Wnt signaling pathway. It also regulates the progression of the cell cycle from the G1 to S phase in many cell types. CCND1 phosphorylates and inactivates retinoblastoma (Rb) protein. Cyclin D1 has been associated with mantle cell lymphoma. This protein is overexpressed in multiple cancers like hepatocellular carcinoma. Cyclin dependent kinase-like 4 (CDKL4) has been associated with various tumors and colorectal cancer. The protein regulates apoptosis of nucleated cells by modifying transcriptional regulation of genes governing cell proliferation and cell death. Like CCND1, it also modulates the progression of cycle from G1 to S phase.
