Gene Information

human ... TPO(7173)

General description

Thyroid peroxidase (TPO) is a membrane-bound, glycosylated, heme-containing peroxidase. It is encoded by the gene mapped to human chromosome 2p25.3. The encoded protein is a member of the human peroxidases family. TPO is a ~100 kDa protein with 933-amino acids. Mature TPO is characterized with a short transmembrane and cytoplasmic regions, as well as a dominant ectodomain, containing MPO-like, complement control protein (CCP)-like and epidermal growth factor (EGF)-like domains.

Immunogen

Peptide with sequence C-TRHVIQVSNEVVTDD, from the internal region of the protein sequence according toNP_000538.3; NP_783650.1; NP_783652.1; NP_783653.1

Biochem/physiol Actions

Thyroid peroxidase (TPO) is involved in the biosynthesis of thyroid hormones T3 and T4. It catalyzes the oxidation of the inorganic iodide and coupling of iodinated tyrosine to produce thyroxine and triiodothyronine. Mutation in the gene increases the risk of susceptibility to autoimmune thyroiditis in patients with allergic rhinitis and asthma. Genetic deficiency of TPO is associated with the development of congenital hypothyroidism.
