General description

Sigma′s denaturing solution is a strong base that denatures double-stranded DNA so that it can be efficiently transferred to nylon membrane and subsequently hybridized to labeled probe. When used with a depurination solution, treatment results in the hydrolysis of the phosphodiester backbone at the sites of depurination, allowing for more efficient transfer of large DNA in neutral Southern transfers.

Application

Denaturation Solution is suitable for denaturing dsDNA before the transfer to membranes during Northern and Southern blotting procedures.

Components

The denaturing solutions is composed of 0.5N NaOH, and 1.5M NaCl.
