General description
Exonuclease I.
The EXO1 (exonuclease I) gene is mapped to human chromosome 1q43 and codes for a 5′→3′ exonuclease of 846 amino acids. It belongs to the 5′-nuclease superfamily.
Application
Acts specifically on single-stranded DNA degrading it processively in the 3′- to 5′-direction, producing 5′-mononucleotides.
Biochem/physiol Actions
Exonuclease I (EXO1) maintains genomic stability by participating in DNA mismatch repair system, DNA recombination, DNA replication and DNA double-stranded break repair in eukaryotes. It also modulates meiosis, immunoglobulin maturation, and micro-mediated end-joining. It functions on DNA with blunt, nicked, gapped and 3′-overhangs. It also possesses endonuclease action and removes 5′-single-stranded flaps that protrudes from a double stranded DNA. Mutation in EXO1 gene results in increased susceptibility to develop cancer.
