General description

Superose® 6 Increase 10/300 GL is a versatile, prepacked column for high-resolution size exclusion chromatography in small-scale (< 500 μl sample volume) preparative purification, as well as for characterization and analysis of proteins and other biomolecules with molecular weights between 5 000 and 5 000 000.

Features and Benefits

  • Increased resolution compared with Superose 6, for higher purity and improved analysis results.
  • Reduced runtime compared with Superose 6, for rapid results.
  • Versatile use for both preparative and analytical purposes, especially for large proteins and complexes.
  • Tolerance to repeated harsh cleaning procedures at high pH, giving long column lifetime and minimal carry-over.

Suitability

Stable in all common buffers: 1 M acetic acid, 8 M urea, 6 M guanidine hydrochloride, 1% SDS, organic solvents, 0.5 M NaOH (for cleaning-in-place)
Superose® 6 Increase 10/300 GL column is suitable for small-scale (25 – 500 μl sample volume) preparative purification for obtaining milligram amounts of highly size-homogenous target proteins at the polishing stage of a purification procedure. It is also well-suited for the characterization of large protein complexes and for the study of protein-protein interactions.

Storage and Stability

Store at 4 to 30 °C (20% Ethanol)

Detection of dynamic protein-protein interactions within complexes and networks remains a challenging task. Here, we show by the example of the proteolytic ClpXP complex the utility of combined chemical cross-linking and mass spectrometry (XL-MS) to map interactions within ClpP and

Pressure is generated by the flow through the chromatographic system. For optimal chromatography functionality, it is important to understand the principle of the pressure drop over the different parts of a system.

Protocols

Superose from GE Healthcare are Size Exclusion Chromatography media consisting of a composite base matrix of dextran and agarose. This page shows how to perform a separation with a superose column.

Samples for chromatographic purification should be clear and free from particulate matter. Simple steps to clarify a sample before beginning purification will avoid clogging the column, can reduce the need for stringent washing procedures, and can extend the life of the chromatographic medium.

Our team of scientists has experience in all areas of research including Life Science, Material Science, Chemical Synthesis, Chromatography, Analytical and many others.
