General description

Immersion Oil has a defined refractive index of about 1.516. It is important that the refractive index (nD) is about 1.5, similar to that of glass. This enables a homogeneous oil immersion to be achieved. Immersion media are used in conjunction with immersion objectives and are located between the surface of the specimen and the lens of the microscope. Immersion oils practically eliminate light beam deflection so that the effectiveness of the lens is considerably increased.

Application

Immersion oil has been used in the following applications:
  1. in epifluorescent microscopy and cell identification
  2. in the assembly and filling of the microinjecting system
  3. in microscopy to reliably visualize dendritic spines

Biochem/physiol Actions

Immersion oil has a larger impact on 3D resolution. Specifically, matching the immersion oil refractive index to glass, reliably enhances the ability to visualize dendritic spines.
