Application

The relationship between polyamine systems and cancer---as well as critical normal cell processes--- led to the creation of clickable polyamine derivatives which are innovative and novel tools for functional studies of the polyamine transport system (PTS). Spermidine and other important polyamine derivatives were conjugated to a BODIPY fluorophore through click chemistry.

Features and Benefits

The BioTracker Clickable Polyamine Spermidine Live Cell Probe is click-conjugated to a green fluorophore to enable the study of polyamine tracking with superior cellular uptake. Probe may be dissolved in DMSO to make a 10mM stock solution. 1 uL of a 10 mM stock solution may be added to 1 mL media to produce a 10 uM working solution. Add probe/media solution to washed cells and incubate at 37C for up to 4h.

Target description

Polyamines play a variety of critical roles in the cell. Some of these processes include gene expression, cell proliferation and differentiation, and cellular stress management.

Polyamine dysregulation is key in a variety of cancers. In colorectal cancer, polyamine content and biosynthesis is significantly higher when compared to normal colorectal tissue. A relationship between polyamines and their metabolites has been used to observe progression in cancers of the breast, lung, colorectal system, ovary, prostate, and pancreas.

In normal cells, there is tight regulation of polyamines through normal cellular mechanisms. One of these is the polyamine transport system (PTS). The characteristics of the PTS are not fully understood, but study of the PTS has shown that many cancer cells have high PTS activity, suggesting this system as a promising target for cancer research.
