General description

Rabbit polyclonal antibody supplied as lyophilized, undiluted serum. Recognizes the serotonin (5-HT) transporter protein.
Recognizes the serotonin (5-HT) transporter in hypothalamus raphe nuclei and spinal cord.
This Anti-Serotonin (5-HT) Transporter (602-622) Rabbit pAb is validated for use in Frozen Sections, Immunofluorescence for the detection of Serotonin (5-HT) Transporter (602-622).

Immunogen

Rat
a synthetic peptide corresponding to amino acids 602-622 of rat 5-HT transporter

Application

Frozen Sections (1:10,000-1:15,000; see application references)
Immunoblotting (see comments)
Immunofluorescence (1:200-1:400)

Warning

Toxicity: Standard Handling (A)

Physical form

Undiluted serum.

Reconstitution

Reconstitute the lyophilized antibody with 100 µl of sterile distilled H₂O. Reconstitute the entire contents of the vial; during shipment and handling portions of the lyophilized pellet may have become dislodged and may not be in the bottom of the vial. Following reconstitution, aliquot and freeze (-20°C). Stock solutions are stable for up to 6 months at -20°C. Further dilute with sterile PBS or Tris buffer to dilutions no higher than 1:10.

Analysis Note

Positive Control
Rat naphe nuclei, hypothalamus, cortex, or spinal cord

Other Notes

Antibody specificity was examined in the rat cortex, hypothalamus, raphe nuclei, and spinal cord. Staining is completely eliminated by pretreatment of antibody with Serotonin (5-HT) Transporter Control Peptide (Cat. No. PP87) at a concentration of 5 µg/ml. Low levels of glutaraldehyde (0.1%-0.3%) may be used in conjunction with paraformaldehyde for tissue perfusion fixation. The specificity of this antibody was also determined by immunoblotting analysis using rat brain extracts of the cortex, hypothalamus, midbrain, and hindbrain, which specifically detected a single band. Antibody should be titrated for optimal results in individual systems.
