Specificity

Clone ORF2-2A7 is a rat monoclonal antibody that detects Cilia- and flagella-associated protein 206 (CFAP206). It targets an epitope within 13 amino acids from the C-terminal region.

Immunogen

A linear peptide corresponding to 13 amino acids from the C-terminal region of mouse Cilia- and flagella-associated protein 206 (CFAP206).

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse testis tissue extract.

Western Blotting Analysis: A 1:500 dilution of this antibody detected CFAP206 in Mouse testis tissue extract.

Tested Applications

Western Blotting Analysis: A representative lot detected CFAP206 in Western Blotting application. (Beckers, A., et al. (2020). Development. 147(21):dev188052).

Target description

Cilia- and flagella-associated protein 206 (UniProt: Q6PE87; also known as CFAP206) is encoded by the Cfap206 gene (Gene ID: 69329) in murine species. CFAP206 is an evolutionarily conserved protein that lacks known motifs or domains, except for a unique 280 amino acid domain with conserved motifs GFC and GIL. It localizes to the basal body/ centrosome and to cilia, and its expression correlates with and depends on Foxj1 expression. In vertebrates it plays a role during motile cilia assembly, and its deficiency can lead to abnormal ciliary beating and decreased sperm motility. It is reported to be a potential component of the nexin-dynein regulatory complex and interacts with radial spoke protein 3. Two isoforms of CFAP206 have been described that are produced by alternative spicing. Isoform 1 is expressed in the sperm, oviduct, lung, nasal cavity, brain ependyma, and choroid plexus. This isoform is shown to be essential for sperm motility and is involved in the regulation of the beating frequency of motile cilia on the epithelial cells of the respiratory tract and it is required for the establishment of radial spokes in sperm flagella. Isoform 2 that is expressed in the sperm, oviduct, lung, nasal cavity, brain ependyma, and choroid plexus is also required for sperm motility and is involved in the regulation of the beating frequency of motile cilia on the epithelial cells of the respiratory tract. Clone ORF2-2A7 specifically detects isoform 1 and detects a sequence within 13 amino acids from the C-terminal region, which is missing in isoform 2. (Ref.: Beckers, A., et al. (2020). Development. 147(21); dev188052).
