General description

Membrane cofactor protein (UniProt P15529; also known as Antigen identified by monoclonal antibody TRA-2-10, CD46 antigen, Complement membrane cofactor protein, Measles virus receptor membrane cofactor protein, TLX, Trophoblast leukocyte common antigen, Trophoblast-lymphocyte cross-reactive antigen) is encoded by the CD46 (also known as MCP, TLX, AHUS2, MIC10, TRA2.10) gene (Gene ID 4179) in human. Signal 1-34, Membrane cofactor protein (MCP) or CD46 is a widely expressed complement regulator that inhibits all three pathways of complement activation, classical (CP), alternative (AP), and lectin (LP). It binds to both C3b (AP) and C4b (CP) and acts as a cofactor for their inactivation by the C3b/C4b inactivator enzyme factor I. CD46 is initially produced with an N-terminal signal sequence (a.a. 1-34), which is post-translationally removed to yield the type 1 membrane bound CD46 consisting of a large extracellular (a.a. 35-343) portion, followed by a single transmembrane α-helical region (a.a. 344-366) and a cytoplasmic tail (a.a. 367-392) with several variant structures. The extracellular portion is composed of four extracellular Complement Control Protein (CCP) domains carrying the cofactor activity, a collar STP-rich region, and a short region of unknown function (U). CD46 also acts as a receptor for certain pathogens such as the measles virus (Edmonston strain), human herpesvirus 6, and Neisseria gonorrhoeae. CD46 is also implicated to have a role in human egg fertilization. Several mutations in the CCPs and the transmembrane region of CD46 are linked to atypical hemolytic uremic syndrome (aHUS).

Specificity

Expected to react with isoforms A (no del, ABC1) and isoform B (del 13, ABC2). Reactivity toward other spliced isoforms (Uniprot P15529-3 through P15529-16) has not been determined.

Immunogen

mIg Fc-tagged recombinant protein corresponding to the extracellular domain of human CD46.

Application

Immunocytochemistry Analysis: 4.0 µg/mL from a representative lot detected CD46 in HeLa and A431 cells.
Flow Cytometry Analysis: 0.1 µg from a representative lot detected CD46 in 1X10E6 HeLa cells.
This Anti-CD46 Antibody, clone 8A9.1 is validated for use in Western Blotting, Immunocytochemistry and Flow Cytometry for the detection of CD46.

Quality

Evaluated by Western Blotting in human CD46 extracellular domain mIg Fc fusion protein.

Western Blotting Analysis: 0.2 µg/mL of this antibody detected CD46 in 0.25 µg of human CD46 extracellular domain mIg Fc fusion protein.

Target description

43.7 kDa (isoforms A; no del ABC1) and 44.2 kDa (isoforms B; del 13 ABC2) calculated.
